What output device / Port should I use?

Generally the Com 1 is reserved for an actual Serial connection. If you are using a USB Cable please check to be sure you have installed the USB drivers that came with your plotter. If you have you should find another com port available in the device pull down - usually com3 or com 4, That can be confirmed by navigating to the windows device manager

Right Click on My Computer
Choose Properties
Click the hardware tab
Click Device Manager
Look for ports
Click the plus sign next to ports
Find a port labeled USB/Serial

One you have found the correct port number go back to SignCut and choose that in device manager.

If this does not help please let us know what ports you find in device manager.
